While it has often been assumed that, in humans, synonymous mutations would have no effect on fitness, let alone cause disease, this position has been questioned over the last decade. There is now considerable evidence that such mutations can, for example, disrupt splicing and interfere with miRNA binding. Two recent publications suggest involvement of additional mechanisms: modification of protein abundance most probably mediated by alteration in mRNA stability1 and modification of protein structure and activity,2 probably mediated by induction of translational (...) pausing. After eight pints of beer, he was drunk enough to decide to drop a brick from the overpass into traffic to see if he could hit something; unfortunately, he was not so drunk that he missed. The brick crashed through the windshield on the driver's side (...) of a truck. It hit the driver, Michael Little, in the chest, triggering a fatal heart attack. He stayed conscious long enough to pull the truck safely to the side of the road, thereby perhaps saving other motorists; then he died. Why do we find agents less blameworthy when they face mitigating circumstances, and what does this show about philosophical theories of moral responsibility? We present novel evidence that the tendency to mitigate the blameworthiness of agents is driven both by the perception that they are less normatively competent—in particular, less able to know that what they are doing is wrong—and by the perception that their behavior is less attributable to their deep selves. Consequently, we argue that philosophers cannot rely on (...) the case strategy to support the Normative Competence theory of moral responsibility over the Deep Self theory. However, we also outline ways in which further empirical and philosophical work would shift the debate, by showing that there is a significant departure between ordinary concepts and corresponding philosophical concepts, or by focusing on a different type of coherence with ordinary judgments. The questions and methods of molecular biology and evolutionary biology are clearly distinct, yet a unified approach can lead to deep insights. Unfortunately, attempts to unify these approaches are fraught with pitfalls. In this informal series of questions and answers, we offer the mechanistically oriented biologist a set of steps to come up with evolutionarily reasonable and meaningful hypotheses. We emphasize the critical power and importance of carefully constructed null hypotheses, and we illustrate our ideas with examples representing a range (...) of topics, from the biology of aging, to protein structure, to speciation, and more. We also stress the importance of mathematics as the lingua franca for biologists of all stripes, and encourage mechanistic biologists to seek out quantitative collaborators to build explicit mathematical models, making their assumptions explicit, and their logic clear and testable. Biologists in all realms of inquiry stand to gain from strong bridges between our disciplines. (shrink)

Patients have received experimental pharmaceuticals outside of clinical trials for decades. There are no industry-wide best practices, and many companies that have granted compassionate use, or ‘preapproval’, access to their investigational products have done so without fanfare and without divulging the process or grounds on which decisions were made. The number of compassionate use requests has increased over time. Driving the demand are new treatments for serious unmet medical needs; patient advocacy groups pressing for access to emerging treatments; internet platforms (...) enabling broad awareness of compelling cases or novel drugs and a lack of trust among some that the pharmaceutical industry and/or the FDA have patients’ best interests in mind. High-profile cases in the media have highlighted the gap between patient expectations for compassionate use and company utilisation of fair processes to adjudicate requests. With many pharmaceutical manufacturers, patient groups, healthcare providers and policy analysts unhappy with the inequities of the status quo, fairer and more ethical management of compassionate use requests was needed. This paper reports on a novel collaboration between a pharmaceutical company and an academic medical ethics department that led to the formation of the Compassionate Use Advisory Committee. Comprising medical experts, bioethicists and patient representatives, CompAC established an ethical framework for the allocation of a scarce investigational oncology agent to single patients requesting non-trial access. This is the first account of how the committee was formed and how it built an ethical framework and put it into practice. (shrink)

Discussions of the Tylenol and Exxon Valdez cases found in textbooks, public relations scholarship, and news coverage are assessed to understand the meanings that practitioners, educators, critics, and journalists have attributed to those events. The essay objects to a central claim made by critics who say these cases set standards for ethical behavior in public relations. This claim, according to us, mistakes moral drama for ethical deliberation.

Formalised knowledge systems, including universities and research institutes, are important for contemporary societies. They are, however, also arguably failing humanity when their impact is measured against the level of progress being made in stimulating the societal changes needed to address challenges like climate change. In this research we used a novel futures-oriented and participatory approach that asked what future envisioned knowledge systems might need to look like and how we might get there. Findings suggest that envisioned future systems will need (...) to be much more collaborative, open, diverse, egalitarian, and able to work with values and systemic issues. They will also need to go beyond producing knowledge about our world to generating wisdom about how to act within it. To get to envisioned systems we will need to rapidly scale methodological innovations, connect innovators, and creatively accelerate learning about working with intractable challenges. We will also need to create new funding schemes, a global knowledge commons, and challenge deeply held assumptions. To genuinely be a creative force in supporting longevity of human and non-human life on our planet, the shift in knowledge systems will probably need to be at the scale of the enlightenment and speed of the scientific and technological revolution accompanying the second World War. This will require bold and strategic action from governments, scientists, civic society and sustained transformational intent. (shrink)

All rights reserved.We present new observations of the low-mass companion to HD 984 taken with the Gemini Planet Imager as a part of the GPI Exoplanet Survey campaign. Images of HD 984 B were obtained in the J and H bands. Combined with archival epochs from 2012 and 2014, we fit the first orbit to the companion to find an 18 au orbit with a 68% confidence interval between 14 and 28 au, an eccentricity (...) of 0.18 with a 68% confidence interval between 0.05 and 0.47, and an inclination of 119°with a 68% confidence interval between 114°and 125°. To address the considerable spectral covariance in both spectra, we present a method of splitting the spectra into low and high frequencies to analyze the spectral structure at different spatial frequencies with the proper spectral noise correlation. Using the split spectra, we compare them to known spectral types using field brown dwarf and low-mass star spectra and find a best-fit match of a field gravity M6.5 ±1.5 spectral type with a corresponding temperature of K. Photometry of the companion yields a luminosity of log=2.88 ± 0.07 dex with DUSTY models. Mass estimates, again from DUSTY models, find an age-dependent mass of 34 ±1 to 95 ±4 M Jup. These results are consistent with previous measurements of the object. (shrink)

Celem artykułu jest przeanalizowanie specyficznego rozumienia współczesnego ateizmu, który 1) powstał we francuskim środowisku filozoficznym w XX i XXI wieku; 2) łączy w sobie krytykę religii (mniej czy bardziej radykalną) z pozytywną propozycją zastąpienia tradycyjnej duchowości religijnej duchowością ateistyczną. Celem badań nad taką duchowością jest weryfikacja następujących hipotez badawczych: 1) „nowa duchowość” jest propozycją alternatywną dla tradycyjnych „duchowości religijnych”, adresowaną do ludzi o przekonaniach ateistycznych; 2) nowa „świecka duchowość” w wielu aspektach jest zbieżna z duchowością religijną, zasadnicza różnica pojawia się (...) w kwestiach rozumienia nieśmiertelności człowieka i sposobu przezwyciężania lęku przed śmiercią; 3) „duchowość ateistyczna”, jako uzupełnienie krytyki postaw religijnych, może przyczynić się do konwersji osób tradycyjnie religijnych i przyjęcia przez nich duchowości, która nie odwołuje się do osobowej Transcendencji. French atheist sprituality The purpose of the article is the analysis of a specific understanding of contemporary atheism, which 1) arose in the French philosophical circles in the 20th and 21st century; 2) combines a critique of religion (more or less radical) with a positive proposal of replacing traditional religious spirituality with an atheist spirituality. The goal of this research is to verify the following hypotheses: 1) a “new spirituality” is an proposal alternative to traditional “religious spiritualities”, addressing atheits; 2) this new “secular spirituality” is in many aspects similar to religious spirituality, with its difference being the new ways of understanding immortality and the ways of overcoming the fear of death; 3) “atheist spirituality”, as a support for the criticque of religious positions, may encourage traditionally religious people to accept a spirituality that requires no relation to personalized Transcendence.Research will be conducted on the base of a critical analysis of the sources of four French thinkers: M. L’expression « langue de bois » y a gardé une tout autre gravité et l’utilisation banalisée qui se pratique (...) à l’Ouest dénote plutôt une incompréhension ou une ignorance de ce qu’est véritablement la langue de bois. Pour ceux qui l’ont combattue, il y a une différence de nature et non pas de degré entre la langue vive et la langue de bois, et c’est que nous voudrions illustrer brièvement, en rappelant la force des protestations à son égard, puis en essayant d’éclairer ce qui a été l’enjeu d’une telle protestation, en nous appuyant principalement sur l’exemple de la Pologne.The French expression « langue de bois » – literally « wooden language » but often referred to as « doublespeak » in English – originated in Eastern Europe. In its current usage in France, however, it has come to cover a range of meanings and refers to any kind of stereotyped discourse, serving as a mere code-word for public speaking, and is often confused with « politically correct ». In Eastern Europe, which has seen totalitarianism of two kinds, this semantic weakening is regarded with dismay. In these countries, the expression – derived from the Russian « yezik douba », literally « tongue of oak » – has lost none of its weight of meaning, and its offhand use in the West is perceived as equating with a lack of understanding and even ignorance of what it really means. To those who fought against it, there is a difference not only in degree but actually in nature between a living language and the « wooden language » of officialdom. This is what we would like to illustrate, briefly, first through the strength of the protests against it and secondly by attempting to shed some light on what the protest was really about, mainly through the example of Poland. L'existence d'une communauté de valeursbasée principalement sur un consensus autour de la culture politique partagée fait office de certitudes dont la remise en cause s'apparente à un pêché contre le code moral moderne qui est celui de l'ordre démocratique et des droits de l'homme. Or, l'Europe est le lieu par excellence de la diversité et des conflits (...) qui en résultent et qui ont souvent été sanglants. Toute cette dimension de confrontation de paradigmes culturels, de l'altérité difficilement tolérée, de la difficile négociation entre ce qui unit et ce qui sépare est souvent occultée dans la communication sur l'Europe. Une approche interdisciplinaire, puisant dans les recherches de l'anthropologie culturelle, de la sociologie des relations interethniques et de la communication interculturelle semble plus à même de répondre à ce type de questionnements nécessaires à l'heure de la crise de la construction européenne.Public debate about the European constitutional treaty provided an opportunity to discuss common values that are ideals for all Europeans. Any criticism of the existence of a community based on a consensus constructed around a certain shared political model is not allowed. In fact, such criticism would be seen as a sin against the modern moral code which is based on a democratic order and the respect of human rights. The problem is that Europe is a place of diversity and, often, of violent conflicts. This aspect of cultural confrontation, lack of tolerance for otherness and the difficulty in negotiating what unites and what divides nations very often remains hidden in public communication about Europe. An interdisciplinary approach, inspired by cultural anthropology, inter-ethnic relations, sociology and cross-cultural communication, is more adapted to analyze such problems which need to be dealt with now while European integration is underway. The result here is a presentation which is inspired and meaningful itself, astonishing its contemporaries and impressing its deseendants. Now, thirty years later, anthropology and psychology have passed beyond L~vy-Bruhl and Spranger. The moderate idealist interpretation of religion as it is found in Parts IV and V, "The World" and "Forms," must give way to descriptive phenomenology and existential analysis, (...) taking into consideration the internal dialectics of religion as well as the dialectics between the religious and the secular. The domain of meaning (Sinnesreich) appears, upon closer consideration, to be a function of the domain of existence. Contemporary religious consciousness and the confrontation of different such consciousnesses would require a "Part Six." The necessarily brief methodology, attached as Epilegomeua to the substance of the book, only contains the remnants of phenomenological epistemology. Besides at least making the main work of Van der Leeuw more known to the Englishspeaking world, this edition may also stimulate phenomenological discussion where Geisteswissenscha /t has a different connotation than it does in continental Europe. In this discussion, several points will have to be made. Is this book a pure phenomenology, with its evaluative character, its tendency to psychological instead of philosophical foundation, and its lack of philosophy of human consciousness? It should be mid that, unfortunately, hardly any phenomenologist of religion has been philosophically trained. So questions like those of the relation between phenomenology and history, the nature of phenomenological objectivity, the premisses and limits of phenomenological understanding, not to say theological implications of any phenomenology of religion, remain open. Van der Leeuw himself, as a scholar and a man, touched on them, but they still have to be worked out. JACQUESWAARDENBURG University of California, Los Angeles Systematic Theology, III. By Paul Tillich. (Chicago, University of Chicago Press, 1963. Pp. 434. $6.95.) The publication of Tillich's Systematic Theology, Volume III, brings to completion the enterprise begun with Volume I in 1951 and Volume II in 1957. As the author tells us, the project of constructing a systematic theology began as far back as the 1920's at the University of Marburg. Suspended when Tillich fled Hitler's Germany in 1933, it was continued during his twenty years at Union Seminary and his years as a university professor at Harvard. Thus in a significant sense, Volume III completes the work of a lifetime. Yet in another equally important sense it marks the permanent incompleteness of this work. For while this is systematic theology in the sense that it acknowledges and works in terms of rationality or coherence, it is clearly not a system in any final, closed, or absolute sense. Not only does Tillich expressly deny any such aspiration, but his openness to facts and to other men's viewpoints, his mixture of humility and candor in the presence of large and difficult problems all make clear the validity of this denial. It is perhaps worth noting in the present context that at no point in Volume III or indeed throughout the whole Systematic Theology does Tillich appeal to any criterion of judgment other than reasoning from facts. At no point is the reader asked to accept any judgment "on faith" or on any extra-rational authority. To be sure, faith is a basic category for Tillich and a crucial one for the Systematic Theology. Indeed many pages are devoted to its exposition and explication, but never is it understood or used as a basis for accepting propositions beyond or against the evidence. Indeed that whole view of faith is abhorrent to Tillich, and a major theme of his work has been to seek an understanding of faith more adequate alike religiously and intellectually. Among contemporary theologians Tillich is preeminent for the seriousness of his commitment to philosophy. In an autobiographical sketch written many years ago, he placed himself on the border between philosophy and theology. As we read the present volume, the conclusion is borne home that he has continued to live on that border territory, maintaining BOOK REVIEWS 299 an active and significant relation to both adjoining realms. Dans une approche psycho-développementale et systémique, cet article examine l’expérience grand-parentale en contexte de troubles du spectre autistique ( tsa ) de l’enfant en considérant la pluralité des relations familiales (conjugale, co-grand-parentale, parents/enfant(s), grand(s)-parent(s)/petit(s)-enfant(s), etc.) dans lesquelles les grands-parents sont inscrits. L’analyse des entretiens menés auprès de six grands-parents (quatre familles) montre notamment l’important engagement et le soutien instrumental et émotionnel des grands-parents auprès des parents et de leur petit-enfant porteur de tsa. In the traditions of narrative ethics and casuistry, stories have a well-established role. Specifically, illness narratives provide insight into patients’ perspectives and histories. However, because they tend to see fiction as an aesthetic endeavour, practitioners in these traditions often do not realize that fictional stories are valuable moral sources of their own. In this paper I employ two arguments to show the mutual relationship between bioethics and fiction, specifically, science fiction. First, both discourses use imagination to set a scene and (...) determine a perspective. Second, bioethics and science fiction share the family resemblance of expressing moral beliefs. I then consider how understanding bioethics and science fiction as interrelated discourses can be the basis of a methodology for inquiry into relational autonomy in the context of biotechnologies and medicine. As an example of this methodology, I analyse Fay Weldon’s novel The Cloning of Joanna May. (shrink)
